Rat Model Study on RAD 140 Neuroprotection
In this study, the researchers investigated the neuroprotective effects of RAD140, which belongs to a class of synthetic compounds known as selective androgen receptor modulators. The decline in testosterone levels during aging is associated with an increased risk of dysfunction and disease in androgen-responsive tissues, including the brain. Testosterone therapy has potential risks, such as an increased risk of prostate cancer. SARMs are designed to selectively activate androgen receptors in specific tissues while minimizing effects on others.
The researchers found that RAD140 demonstrated neuroprotective effects both in cultured rat neurons and in the brains of male rats. In cultured neurons, RAD140 was as effective as testosterone in reducing cell death induced by apoptotic insults. The neuroprotective action of RAD140 was dependent on the MAPK signaling pathway. In in vivo experiments with gonadectomized male rats, RAD140 exhibited peripheral tissue-specific androgen action, sparing the prostate while activating androgenic gene regulation effects in the brain. RAD140 also provided neuroprotection to hippocampal neurons against cell death induced by the excitotoxin kainate, a model relevant to neurodegenerative diseases like Alzheimer’s.
Muscle Hypertrophy in Rat Model
This study investigated the potential additive effects of the SARM RAD140 on muscle hypertrophy when combined with functional overloading (FO) of the plantaris muscle in Sprague-Dawley rats. Four treatment groups were established, including RAD140-FO, RAD140-Control, Vehicle-Control, and Vehicle-FO. The rats in RAD140 groups received the drug through drinking water, while control groups received only the vehicle (methylcellulose). Muscle weight analysis revealed a significant increase in RAD140-FO groups compared to control groups, although not statistically different from the vehicle-FO group. Similar findings were observed in muscle fiber cross-sectional area (CSA). The results suggest the potential of RAD140 to stimulate muscle hypertrophy, but further investigation with longer treatment duration is needed for a better understanding of the physiological response to functional overloading and SARMs treatment.
