Naturally produced Kisspeptin peptide plays an important role in the reproductive physiology of mammals. Kisspeptin 10 is often thought to stimulate the release of GnRH from the hypothalamus. By stimulating the release of GnRH from the hypothalamus it stimulates gonadotropin released from the pituitary gland. Being that kisspeptin 10 is a shorter amino acid sequence it makes it much simpler to be digested.
Gametogenic and Steroidogenic Action of Kisspeptin-10 in Asian Catfish
The study aimed to investigate the direct action of kisspeptin-10 (KP-10) and its role in gonadal activities in the gonadally quiescent Asian catfish using both in vivo and in vitro approaches. Unlike mammals, fish possess two kisspeptin genes, kiss1 and kiss2, with varied and contradictory differences in their reproductive activities.
In vivo treatment with KP-10 induced the precocious onset and rapid progression of gametogenesis in Clarias batrachus, as evidenced by advanced stages of ovarian follicles and germ cells in the testis compared to control gonads. KP-10 treatment also elevated steroid levels in both in vivo and in vitro conditions. [3]
It increased the expressions of key steroidogenic enzymes such as 3β-HSD, 17β-HSD, and StAR protein, responsible for cholesterol transfer within steroidogenic cells. Moreover, KP-10 augmented the activities of 3β-HSD and 17β-HSD in ovarian explants and up-regulated the expressions of MAPK components (pERK1/2 and ERK1/2) in gonadal explants. [3]
The findings suggest that kisspeptin-10 stimulates gametogenesis by enhancing gonadal steroid production in the Asian catfish. The study delineates the putative mechanistic cascade of steroidogenic actions of kisspeptin-10 in teleost fish. Additionally, it proposes that kisspeptin may act locally to regulate gonadal activities in an autocrine/paracrine manner, independent of known extra-gonadal factors in the catfish.
Influence of Kisspeptin 10 on Milk Synthesis in Bovine Mammary Epithelial Cells
The study aimed to unravel the mechanism by which Kisspeptin-10 (Kp-10), a peptide hormone, influences milk synthesis in bovine mammary epithelial cells (BMECs), as its role in this process remains unclear. Researchers employed various approaches, including the use of a GPR54 inhibitor, SIRT6 overexpression plasmid, and siRNA, to investigate the regulatory mechanisms of Kp-10 in milk protein and milk fat synthesis.
The findings revealed that treatment with 100 nM Kp-10 significantly increased milk synthesis in BMECs. Notably, SIRT6 overexpression substantially attenuated milk protein and milk fat synthesis in these cells. Furthermore, the overexpression of SIRT6 reversed the activation of the Kp-10-induced mTOR signaling pathway, a key pathway involved in protein synthesis and cellular growth. [4]
Further analysis suggested that SIRT6 might exert its regulatory effect on the mTOR signaling pathway at the transcriptional level. The results strongly indicate that the Kp-10/GPR54 axis activates the mTOR signaling pathway by suppressing SIRT6 expression, consequently promoting milk synthesis in BMECs. [4]
This comprehensive understanding of the molecular mechanisms underlying Kp-10’s regulation of milk synthesis sheds light on potential targets for enhancing milk production and quality in dairy animals.
Kisspeptin Administration on Adiponectin, Leptin, and Resistin Secretion in Adult Male Rhesus Monkey
In recent years, kisspeptin-KISS1R signaling has emerged as a key regulator of reproductive function across various vertebrate species. While traditionally associated with reproductive tissues, both KISS1 (encoding kisspeptin) and its receptor, KISS1R, are expressed in multiple tissues, including adipose tissue. However, the local role of kisspeptin in adipocyte function remains unclear.
In this study, the effects of exogenous human Kisspeptin-10 on three important adipokines – adiponectin, leptin, and resistin – were investigated in intact adult male rhesus monkeys under different feeding conditions: normal fed, 24-hour fasting, and 48-hour fasting. Fasted monkeys exhibited decreased plasma resistin and leptin levels (p<0.01), while adiponectin levels increased (p<0.05). [7]
Kisspeptin administration significantly increased mean plasma adiponectin levels (p<0.05) under fed and 24-hour fasting conditions compared to pretreatment or vehicle-treated levels. A stimulatory effect was also observed on plasma adiponectin levels under 48-hour fasting conditions, although it was not statistically significant. No significant effect of kisspeptin was observed on mean plasma leptin and resistin levels. [7]
These findings reveal a novel role of kisspeptin as a regulator of adipocyte function in higher primates, specifically in stimulating plasma adiponectin levels under fed and 24-hour fasting conditions in adult male rhesus monkeys.
