What is Ipamorelin?

Ipamorelin is a peptide that consists of a five amino acid chain. It’s primarily known as a secretagogue, this means that it promotes secretion of certain hormones. Specifically, Ipamorelin promotes the secretion of growth hormone from the pituitary gland. While doing so, it also works to mimic ghrelin. Ghrelin is the hunger hormone and is created within the confines of the stomach.

Ipamorelin Induces Longitudinal Bone Growth in Rats

Ipamorelin, a potent synthetic pentapeptide, possesses specific growth hormone (GH)-releasing properties. In a study on adult female rats, ipamorelin was injected subcutaneously at different doses three times daily for 15 days to investigate its effects on longitudinal bone growth rate (LGR), body weight (BW), and GH release.

Results showed that ipamorelin dose-dependently increased LGR, with rates rising significantly compared to the control group. There was also a significant and dose-dependent effect on BW gain. However, ipamorelin treatment did not influence total IGF-I levels, IGFBPs, or serum markers of bone formation and resorption. The number of tartrate-resistant acid phosphatase-positive multinuclear cells in the tibia’s metaphysis did not change significantly with treatment. [4]

The responsiveness of the pituitary to a provocative intravenous dose of ipamorelin or GHRH showed a marginal reduction in plasma GH response after ipamorelin, but no change after GHRH. The pituitary GH content remained unaffected by ipamorelin treatment. [4]

Comparative Study of Ipamorelin and GHRP 6 on Bone Mineral Content in Female Rats

The study investigated the effects of growth hormone (GH) secretagogues (GHSs) on bone mineral content (BMC) in young adult female rats. Rats were given ipamorelin (IPA), GH-releasing peptide-6 (GHRP-6), GH, or vehicle continuously via osmotic minipumps for 12 weeks. Dual X-ray absorptiometry (DXA) measurements were taken every 4 weeks, and femurs were analyzed by mid-diaphyseal peripheral quantitative computed tomography (pQCT) scans. Femurs and vertebrae L6 were analyzed using Archimedes’ principle and ash weight determinations.

All treatments increased body weight and total tibial and vertebral BMC measured by DXA compared to controls. However, total BMC corrected for body weight increase remained unaffected. Tibial area bone mineral density (BMD) increased, but total and vertebral area BMDs remained unchanged. pQCT measurements revealed that increased cortical BMC was due to increased cross-sectional bone area, while cortical volumetric BMD remained unchanged. Femur and vertebra L6 volumes increased, but volumetric BMDs were unaffected. Ash weight increased with all treatments, but mineral concentration remained unchanged. [5]

In conclusion, treatment with the GHSs ipamorelin and GHRP-6 increased BMC as measured by DXA in vivo. In vitro measurements using pQCT and Archimedes’ principle, along with ash weight determinations, showed that increases in cortical and total BMC were due to increased bone dimensions, while volumetric BMD remained unchanged. Efficacy of Ipamorelin in Rodent Model of Postoperative Ileus

The study investigated the potential of ipamorelin, a selective growth hormone secretagogue and ghrelin receptor agonist, to improve gastrointestinal transit and alleviate symptoms in a rodent model of postoperative ileus (POI). Male rats subjected to laparotomy and intestinal manipulation were fasted, and a dye marker was infused into the proximal colon to assess postsurgical colonic transit time.

Ipamorelin, along with growth hormone-releasing peptide (GHRP)-6, or saline (vehicle), was administered via intravenous bolus infusion after a single dose or a 2-day repetitive dosing regimen. The effects were evaluated by measuring the time to the first bowel movement, cumulative fecal output, food intake, and body weight over 48 hours post-surgery. [6]

Results indicated that a single dose of ipamorelin or GHRP-6 decreased the time to the first bowel movement compared to the vehicle group. However, it did not significantly affect cumulative fecal output, food intake, or body weight gain after 48 hours. [6]

Conversely, repetitive dosing of ipamorelin significantly increased cumulative fecal pellet output, food intake, and body weight gain. These findings suggest that repeated intravenous infusions of ipamorelin may offer symptomatic relief in cases of postoperative ileus.
