IGF1 LR3 or long arginine 3 IGF 1, is a synthetic peptide. In this form it is referred to as a lengthened analogue of insulin like growth factor one. The main difference is that IGF 1 has a glutamic acid in the third position of its chain. When looking at IGF LR3, long arginine 3 IGF 1 has an arginine acid in the third position instead. It also has 13 more amino acids at its N-terminus. Because of the modifications it is able to retain its pharmacological activity and is three times more potent than IGF 1. The insulin-like growth factor (IGF) system is a complex network involving IGF-I and -II ligands, binding proteins (IGFBP-1 to -6), and receptors, particularly the type I IGF receptor (IGF-I-R). Maternal IGFs, abundantly secreted by the female reproductive tract, likely influence embryonic growth and differentiation through modulation of IGF effects by embryonic IGFBP and IGF-I-R expression.

A study investigated the effects of recombinant IGF-I and IGF LR3 on in vitro produced bovine embryos. IGF LR3, with a much lower affinity to IGFBPs compared to IGF-I, stimulated early embryonic cleavage more effectively, while further development was better supported by IGF-I. Blastocysts’ total cell numbers were highest with IGF LR3, followed by IGF-I and control groups, primarily due to differences in trophectoderm cell numbers. [1]

Messenger RNA (mRNA) expression analysis showed that IGF LR3 treatment increased IGFBP-2 levels significantly compared to IGF-I and control groups. Both IGF treatments elevated IGFBP-5 mRNA levels, with slightly higher levels in the IGF-I group. IGFBP-3 mRNA abundance was higher in IGF-I-treated embryos compared to IGF LR3-treated ones. IGF-I reduced IGF-I-R mRNA levels, while IGF LR3 increased them compared to the control group. [1]

Overall, the affinity of IGF peptides to IGFBPs influences preimplantation embryo development and various parameters such as cell numbers and mRNA expression of components of the IGF system in distinct directions.

Comparison of IGF-1 and IGF LR3 in Rat Model

The study examined the relative potency of IGF-I and the analogue IGF LR3 in promoting growth or reversing catabolism in rats via injection rather than continuous infusion. IGF LR3, with low affinity for IGF-binding proteins in rats, is cleared more rapidly from circulation compared to IGF-I. Experiments were conducted in normal growing rats and those made catabolic by dexamethasone infusion. [2]

Results showed that continuous infusion of IGFs exhibited greater efficacy than injection modes, particularly in anti-catabolic actions. IGF LR3, when continuously infused, was generally 1.5- to 2-fold more potent than IGF-I in changes related to body weight gain, visceral organ weights, and feed use efficiency. Even with once-daily injection, IGF LR3 remained more potent than IGF-I in several effects, including reducing Nτ-methylhistidine excretion by dexamethasone-treated rats. [2]

Despite its faster clearance, injected IGF LR3 maintained superior potency to injected IGF-I for various actions, although the potency was significantly reduced compared to continuous infusion. The study suggests that using IGF analogues with low binding protein affinity may offer advantages in potency and tissue specificity when IGFs are administered via injection.
