Comparison of the Effects of Ghrelin, GHRP-6 and Motilin in Rat Model

The study compared the gastroprokinetic effects of ghrelin, growth hormone releasing peptide 6 (GHRP-6), and motilin in rats both in vivo and in vitro. Ghrelin and GHRP-6, but not motilin, were found to accelerate gastric emptying and transit in rats, with ghrelin demonstrating greater potency than GHRP-6. The dose-response relationship for ghrelin was bell-shaped, unlike GHRP-6.

In antral and fundic strips, neural responses to electrical field stimulation (EFS) included an on-relaxation reversed into a cholinergically mediated contraction by the addition of NG-nitro-l-arginine methyl ester hydrochloride (l-NAME). Ghrelin agonists reduced on-relaxations in fundic strips and increased cholinergic off-contractions in both antral and fundic strips. The concentration-response curves for muscle strips of the fundus were bell-shaped, with maximal effects observed for ghrelin and GHRP-6. [4]

No effects were observed with motilin across the different experimental conditions. The presence of growth hormone secretagogue receptor 1a transcripts in the strip preparations was confirmed by reverse transcriptase polymerase chain reaction (RT-PCR). [4]

In conclusion, ghrelin and GHRP-6 accelerate gastric emptying and transit by activating cholinergic excitatory pathways in the enteric nervous system, in addition to the known vagal pathways. Motilin, however, did not exhibit similar effects under the studied conditions.

Effects GHRP-6 on Septic Ileus in Mice

The study investigated the effects of ghrelin and growth hormone-releasing hormone 6 (GHRP-6) on gastric emptying and transit in both control and septic mice. Mice were administered lipopolysaccharides (LPS) to induce sepsis or saline as a control. Following a 16-17 hour period, mice received pretreatments of saline, ghrelin, or GHRP-6 before intragastric administration of Evans blue dye. Gastric emptying, transit, rectal temperature, and behavioral scale were assessed.

In control mice, both ghrelin and GHRP-6 accelerated gastric emptying, while transit remained unaffected. Septic mice displayed delayed gastric emptying and transit, hypothermia, and worsened behavioral scale scores. [5]

In septic mice, ghrelin accelerated gastric emptying without affecting transit, while GHRP-6 significantly accelerated gastric emptying in a dose-dependent manner without significantly impacting transit. Neither ghrelin nor GHRP-6 affected endotoxin-induced hypothermia or behavioral scale deterioration. [5]

The study suggests that the prokinetic effects of ghrelin and, particularly, GHRP-6 could offer therapeutic potential in treating septic gastric ileus.
