The Italian company was experimenting with the semisynthetic derivatives of the ergot alkaloids at the time of the discovery. These type of alkaloids are considered to be secondary metabolites. The groups of ergot alkaloids are clavines, lysergic acid, and ergopeptines.

After its discovery in 1980, Cabergoline underwent testing. Then in 1991 a published abstract appeared at the Society of Neuroscience meeting. New developments ensued, as the company that developed this substance was acquired twice by two separate entities.

Right before the acquisitions began, things were happening for this new drug. In the Netherlands during 1992, it was approved under the name Dostinex and marketed. Then in between the acquisitions it was approved by the FDA in December of 1996.

The first occurrence of acquisition occurred in 1993. The Italian company was acquired by a company named Pharmacia. Then in 2003, the second acquisition occurred when a common company known as Pfizer acquired it.

Studies with Cabergoline

Micro- & Macroprolactinoma

For starters, what the heck is macroprolactinoma and macroprolactinoma? Well, macroprolactinoma is a non-cancerous tumor that develops in the pituitary gland. When a tumor is larger than 1cm, the condition is then referred to as macroprolactinoma.

Patients between the ages of 17 and 70 that experience macroprolactinoma took part in this study. The goal was to test the effects of Cabergoline as a treatment option. In addition to these forty one individuals another ten patients between the ages of 18 and 53 also took part. These ten individuals were experiencing microprolactinoma.

The basic goal of the study is to see if prolactin levels, restoration of pituitary function and treatment of tumors is a possibility when using this particular substance. Over the course of twenty four months, the patients underwent treatment. Within this time frame, results were very promising.

Of the patients, 31 of the macroprolactinoma patients saw their prolactin levels normalize. In addition to the normalization of their prolactin, they also saw galactorrhea disappear as well. What is galactorrhea? It’s basically a milky discharge from the nipple that isn’t related to typical milk production in the breast.

Individuals also had their non-cancerous tumors shrink in both groups. Some of the tumors even disappeared altogether. For some of the patients, headaches were a constant. Fifteen of the patients saw headaches dissipate while taking part in the study with Cabergoline. However, one of the most notable outcomes of the experiment was the normalization of testosterone for 31 patients.

Prolactin

For those that may not know, prolactin is a hormone that is produced by the pituitary gland. This is a relatively small but powerful area located in the brain. It is what causes breasts to develop and create milk once a baby is born for women. Whether you are a woman or man, prolactin is present in small amounts in your blood.

This is where a study done with one hundred and forty women becomes important. Cabergoline was tested to determine its dose dependency on prolactin levels. These 14o women were divided into four groups and were either administered various doses of the substance or a placebo. This meant that there were three groups of forty women with the final group having only 20 women. Of those in the testing groups, they were administered doses of 0.5mg, 0.75mg, and 1.0mg.

It is important to note that the women in this study delivered their baby. Then, twelve hours later received the first treatment. After treatment, the women were examined at 6 hours, 12 hours, the first five days and the fourteenth day. The main focus of these examinations was milk secretion, pain, breast engorgement, side effects, and prolactin levels.

The main outcomes for the women involved varied based on dose. The women taking 1.0mg of Cabergoline saw a 90.2% reduction in lactation. Whereas, those taking 0.5mg only saw a 45% reduction.
